(possible spoilers)

The Key to Hell was originally owned by Lucifer Morningstar. After he decided to retire and releases all the dawned souls and demons from hell, he locks the Gates of Hell and gives the key to Morpheus/Dream. Morpheus/Dream then gave the key to Duma, who then gave it to Cristopher Rudd, until finally, it came to Elaine Belloc.

In Locke & Key

Back in 2014 Joe Hill stated in an interview that the Locke family will have either created the actual Key to Hell that appears in the DC Sandman and Lucifer universe, or a working duplicate. He then went on to describe the plot of “Hell & Gone” as: “It would be a story of Chamberlain Locke, set in the past, and he would have wandered through Hell, and then the House of Mysteries and the House of Dream, and he would have crossed paths with Morpheus and Lucifer and Death, and a lot of the great characters from Vertigo.”

It is said that the mere possession of the key makes its owner Lord of Hell and allows absolute control over Hell.
The key itself locks and unlocks the Gates of Hell.
